The 2003 analysis covered the 1992-2002 period, and the hot research areas indicated by our top papers list included the association of apolipoprotein E type 4 allele with both familial and sporadic AD, candidate genes for familial AD, studies of amyloid beta production, and clinical trials of tacrine and segeline/alpha-tocopherol for treatment of AD.
Flash forward almost 20 years from the 1992 starting point of the original analysis, and the notable research areas picked up by the top papers list for 2000-2010 include the potential for amyloid beta peptide antibody vaccines as a treatment for AD in several murine models and one human case study, the observation and that statin drugs appear to affect amyloid beta peptide levels, the ability to image amyloid levels in the living brain with the tracer Pittsburgh Compound B, and the implications of plasma homocysteine levels as a risk factor for dementia and AD.
Other topics of interest in the past decade include the aforementioned study by Hebert et al. on AD prevalence using US census data, in vivo and in vitro disease models, and pinning down early events in AD, such as mild cognitive impairment and oxidative damage. The amyloid hypothesis, as well as the roles of genes and inflammation in AD, is extensively reviewed.
