In these more recent papers, the concept of the biomarker Pittsburgh Compound B really takes off: three studies examine its efficacy and one further study uses it as a tool to measure fibrillar amyloid beta burden in at-risk cognitively normal adults. Genome-wide association studies also gain quite a bit of attention, as the top two papers discuss variants at CLU, PICALM, and CR1 associated with AD.
Other topics garnering citation attention in this two-year time span include the notion of cortical hubs playing a role in the pathology of AD, the neuroprotective effects of brain-derived neurotrophic factor in rodent and primate models of AD, and impaired balance of mitochondrial fission and fusion in AD. Prospective clinical studies examining the role of statins on amyloid beta levels and diet and exercise on risk of AD are also reported.
Although apolipoprotein E was not mentioned at all on the 10-year list of papers, it comes up more than once on the two-year list in reviews that mention the potential for therapies based on advances in knowledge about apolipoprotein E. Other topics under review include amyloid beta, neurovascular mechanisms and the blood-brain barrier, and the state of the science in AD in 2009.
