Clinical trials dominate both paper lists. The most prevalent therapeutic combination being studied is radiotherapy with concomitant and adjuvant temozolomide, which appears to impart a survival benefit with minimal toxicity. Other therapies being heavily cited over the past decade include EGFR kinase inhibitors, AZD2171, gefitinib, temsirolimus, and bevacizumab plus irinotecan. A multivariate analysis on prognosis, extent of resection, and survival, as well as a population-based study on the frequency of genetic alterations in glioblastoma and their effects on survival, are also included.
Studies at the molecular level receiving citation attention over the past 10 years include integrated genomic analyses to characterize and identify useful genetic alterations, such as aberrant expressions in microRNA, the potential use of CD133 as a marker for chemoresistance, and the role of signal transduction pathways in cell proliferation. The idea that glioblastoma cells have stem-cell-like neural precursors gets a lot of play in the literature. The aims and benefits of the Cancer Genome Atlas pilot project are also addressed.
